Flow cytometric analysis of Rat CD31 expression on SD Rat splenocytes. SD Rat splenocytes were stained with Allophycocyanin Mouse Anti-Rat CD45RA Antibody and either Alexa Fluor® 488 Mouse IgG1, k Isotype Control (left panel) or SDT Alexa Fluor® 488 Mouse Anti-Rat CD31 Antibody (right panel) at 2 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD31 (PECAM-1) is a 130 kDa type I transmembrane glycoprotein of the immunoglobulin superfamily, expressed constitutively on endothelial cells, platelets, monocytes, neutrophils and some T cells, where it localizes to intercellular junctions and functions as a mechanosensitive adhesion and signaling hub that orchestrates leukocyte transendothelial migration through homophilic CD31-CD31 interactions and heterophilic binding to partners such as CD177 and integrins, while simultaneously regulating endothelial permeability, angiogenic sprouting and vascular integrity via Src- and PI3K-mediated phosphorylation of its short cytoplasmic tail, making it a key biomarker for microvessel density in tumors and a checkpoint regulator of monocyte FcγR responses.
