Flow cytometric analysis of Human TCR γ/δ expression on human PBMC. Human PBMC were stained with Brilliant Violet 421™ Mouse Anti-Human CD3 Antibody and either Alexa Fluor® 488 Mouse IgG1, k Isotype Control (left panel) or SDT Alexa Fluor® 488 Mouse Anti Human TCR y/ð Antibody (right panel) at 5μl/test. Total viable cells, as determined by Fixable Viability Dye 662 (S0D0016), were used for analysis. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.

Background
TCR γδ protein is a transmembrane disulfide-linked heterodimer composed of a γ and a δ chain, each containing a membrane-distal immunoglobulin-like variable (V) domain, a membrane-proximal constant (C) domain, a short stalk, a hydrophobic transmembrane anchor and a cytoplasmic tail that associates with CD3 and ζ signaling subunits; unlike conventional αβ T-cell receptors it recognizes native antigens—small pyrophosphate metabolites, lipids presented on MHC-like butyrophilin molecules, stress-induced MIC-A/B or even unprocessed surface proteins—without requiring peptide-MHC, triggering rapid innate-like cytokine and cytotoxic responses that bridge innate and adaptive immunity.
