Background

CD200R is a trans-membrane glycoprotein of the immunoglobulin superfamily encoded by CD200R1, expressed chiefly on myeloid cells (macrophages, dendritic cells, microglia) and some lymphoid subsets; when engaged by its ligand CD200 (OX-2) it becomes tyrosine-phosphorylated in its cytoplasmic NPxY motif, recruits the adaptor Dok-2 and RasGAP, thereby dampening Ras/MAPK and NF-κB signaling to suppress production of TNF-α, interferons and iNOS, curb mast-cell degranulation, restrain ILC2 effector cytokines, and broadly down-regulate innate and adaptive inflammatory responses, a signaling axis that is exploited by tumors for immune evasion and is being therapeutically targeted to treat asthma, neuroinflammation and cancer .

Flow cytometric analysis of human CD200R expression on human peripheral blood cells. Human peripheral blood cells were stained with Alexa Fluor® 488 Mouse IgG1, κ Isotype Control (left panel) or SDT Alexa Fluor® 488 Mouse Anti-Human CD200R Antibody (right panel) at 5 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.
