Background

CD117, also known as c-KIT or stem cell factor receptor, is a 145-kDa type III transmembrane receptor tyrosine kinase encoded by the KIT proto-oncogene on chromosome 4q12; upon binding its ligand stem cell factor (SCF) it dimerizes and autophosphorylates, triggering downstream RAS-MAPK, PI3K-AKT, and JAK-STAT cascades that orchestrate melanogenesis, hematopoiesis, mast-cell development, gametogenesis, and interstitial cell of Cajal function, and gain-of-function mutationsmost commonly in exons 9, 11, 13, or 17drive constitutive kinase activity underlying gastrointestinal stromal tumors (GIST), systemic mastocytosis, and subsets of acute myeloid leukemia, making CD117 a key diagnostic immunohistochemical marker and therapeutic target for tyrosine kinase inhibitors such as imatinib, sunitinib, and avapritinib.

Human PBMC were stained with APC Mouse Anti-Human Lineage Cocktail Antibody and either Alexa Fluor® 488 Mouse IgG1, κ Isotype Control (left panel) or SDT Alexa Fluor® 488 Mouse Anti-Human CD117 Antibody (right panel) at 5 μl/test. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.
