Background

CD107b, also known as LAMP2 (Lysosome-Associated Membrane Protein 2), is a heavily glycosylated type I transmembrane protein that constitutes a major component of the lysosomal membrane, where it plays critical roles in maintaining lysosomal integrity, autophagy, and chaperone-mediated autophagy by acting as a receptor for chaperone–substrate complexes on the lysosomal surface.

Human PBMC (human peripheral blood mononuclear cells) were fixed with 4% PFA and permeabilization with 0.1% Tween. Cells were then intracellularly stained with Brilliant Violet 421™ Mouse Anti-Human CD3 Antibody and either Alexa Fluor® 488 Mouse IgG1, k Isotype Control (left panel) or SDT Alexa Fluor® 488 Mouse Anti-Human CD107b/LAMP2 Antibody (right panel) at 5μl/test. Total viable cells, as determined by Fixable Viability Dye 662 (S0D0016), were used for analysis. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.
