Background

CD107a, also termed lysosome-associated membrane protein-1 (LAMP-1), is a heavily glycosylated type-I transmembrane glycoprotein encoded by the LAMP1 gene, constituting ~50 % of all proteins in the lysosomal membrane where its dense carbohydrate coat protects against self-digestion, while in immune cells it functions as a universally accepted surrogate marker of cytotoxic degranulation because, upon NK- or CD8⁺ T-cell activation, the lytic granules fuse with the plasma membrane and transiently expose CD107a on the cell surface, enabling flow-cytometric quantification of perforin/granzyme release; beyond immunology, CD107a shuttles between lysosomes, endosomes and the plasma membrane, provides carbohydrate ligands for selectins to facilitate leukocyte adhesion, and its overexpression on tumor cells correlates with enhanced metastatic potential .

Flow cytometric analysis of Human CD107a expression in Jurkat cells. Cells from Jurkat (Human T cell leukemia T lymphocyte) was fixed with 4% paraformaldehyde and permeabilized with 90% methanol and then stained with either Alexa Fluor® 488 Mouse IgG1, κ Isotype Control (Black line histogram) or SDT Alexa Fluor® 488 Mouse Anti-Human CD107a antibody (Red line histogram) at 1.25 μl/test, cells without incubation with primary antibody and secondary antibody (Blue line histogram) was used as unlabelled control. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.
