Flow cytometric analysis of Foxp3 / Transcription Factor Staining Buffer Set fixed and permeabilized Human PBMC (human peripheral blood mononuclear cells) were stained with Alexa Fluor® 488 Mouse IgG1, k Isotype Control (black line histogram) or SDT Alexa Fluor® 488 Mouse Anti Human Aiolos antibody (red line histogram) at 5μl/test. Total viable cells, as determined by Fixable Viability Dye 452 (S0D0021), were used for analysis. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
Aiolos (IKZF3) is a lymphoid-restricted zinc-finger transcription factor that partners with Ikaros to control B-cell differentiation, proliferation and apoptosis by remodeling chromatin and directly regulating genes such as BCL2 and BCL6; its expression peaks in peripheral blood leukocytes, spleen and thymus, is induced by TGFB1 and dioxin, and its loss-of-function or deletion is linked to B-cell lymphoma, chronic lymphocytic leukemia and multiple myeloma, while over-expression promotes migration and invasion of lung cancer cells and is being exploited as a target for PROTAC degraders.
