Background

ALDH1A2, also known as retinaldehyde dehydrogenase 2 (RALDH2), is a critical cytosolic enzyme belonging to the aldehyde dehydrogenase superfamily that serves as the primary catalyst for the irreversible oxidation of retinaldehyde into retinoic acid (RA), a potent derivative of vitamin A essential for embryonic development and tissue homeostasis. By tightly regulating local RA concentrations, ALDH1A2 orchestrates fundamental biological processes including anterior-posterior axis patterning, limb bud formation, heart morphogenesis, and the differentiation of various cell lineages through the activation of nuclear retinoic acid receptors (RARs) which modulate gene transcription. Beyond its pivotal role in development, this protein is implicated in adult physiological functions such as immune regulation and tissue repair, while its dysregulation or specific polymorphisms have been linked to congenital malformations, certain cancers where it may influence stemness and drug resistance, and inflammatory conditions, making it a significant target for therapeutic intervention and a key biomarker in developmental biology and pathology research.

Immunocytochemistry

ICC shows positive staining in K562 cells (top panel) and negative staining in Jurkat cells (below panel). Anti-ALDH1A2 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
