Background

AKT1S1, also commonly known as PRAS40 (proline-rich AKT substrate of 40 kDa), serves as a critical "molecular brake" within cells. Its core function is to act as an inhibitory component of the mTORC1 signaling complex, precisely regulating cell growth and metabolism. In the absence of nutrient signals, AKT1S1 binds to the mTORC1 complex, directly blocking the substrate-binding site and thereby suppressing mTORC1 kinase activity, preventing energy-consuming processes such as protein synthesis. Upon stimulation by insulin or growth factors, upstream AKT kinase phosphorylates AKT1S1 at threonine 246 (Thr-246). The phosphorylated AKT1S1 then binds to 14-3-3 proteins and dissociates from the mTORC1 complex, effectively "releasing the brake" to activate mTORC1 signaling and promote cell growth and proliferation. As a key node connecting the PI3K-AKT and mTORC1 signaling pathways, dysregulation of AKT1S1 function is closely associated with various diseases, including cancer and metabolic disorders, and it also plays important roles in stage-specific regulation of normal physiological processes such as spermatogenesis.
