Background
AKAP4, the most abundant protein of the sperm fibrous sheath, is a testis-enriched scaffolding protein that simultaneously binds PKA via dual-specificity anchoring sites and PKC through polybasic motifs, thereby acting as a cAMP/PKA ↔ PKC/ERK1/2 switch that is phosphorylated on Ser/Tyr during capacitation to direct the kinase cascade to the flagellar principal piece, mid-piece and post-acrosomal region, enabling progressive motility and male fertility while also moonlighting in multiple cancers where its cytoplasmic/membrane accumulation drives proliferation, EMT and metastasis through the Shh and Wnt/β-catenin pathways.
