Background
AK1C3 (full name Aldo-keto reductase family 1 member C3, gene name AKR1C3) is a human cytosolic NADPH-dependent oxidoreductase belonging to the aldo-keto reductase superfamily and composed of 323 amino acids. It plays a central role in the metabolism of steroid hormones and prostaglandins, possessing 3α-, 17β-, and 20α-hydroxysteroid dehydrogenase activities that enable it to catalyze the bidirectional interconversion between ketones and alcohols. Specifically, the most notable catalytic function of AK1C3 is the efficient reduction of androstenedione to testosterone; it can also convert estrone to estradiol, reduce prostaglandin D2 to 11β-prostaglandin F2α, and convert the potent androgen dihydrotestosterone (DHT) to the less active 3α-androstanediol, thereby regulating the local availability of androgens, estrogens, and progestogens. The protein is encoded by a gene located at chromosome 10p15.1 and is expressed in a variety of tissues. Dysregulation of its expression is closely associated with the development and progression of multiple malignant tumors, including prostate cancer, breast cancer, lung cancer, and renal cell carcinoma, making it a potential target of considerable interest in cancer therapy research.
