Background

ADORA1 protein (adenosine A1 receptor) is a cell-surface integral membrane protein of 326 amino acids with a molecular weight of approximately 37 kDa, encoded by the human ADORA1 gene. It belongs to the G protein-coupled receptor (GPCR) family and has a typical seven-transmembrane structure. As the subtype with the highest affinity for adenosine among the four adenosine receptor subtypes (A1, A2A, A2B, and A3), ADORA1 is widely expressed in the central nervous system and inhibits adenylyl cyclase activity by coupling to Gi/o family heterotrimeric G proteins, thereby reducing intracellular cAMP levels. This receptor regulates a variety of physiological functions, including heart rate and myocardial contractility, smooth muscle tone, sedation, neurotransmitter release, platelet function, lipolysis, renal function, leukocyte function, and sperm capacitation. ADORA1 is also involved in processes such as signal transduction, apoptosis, phagocytosis, inflammatory responses, and nervous system development, and it co-localizes with adenosine deaminase, which may be required for high-affinity agonist binding. In addition, ADORA1 can form homo- or hetero-oligomers with other adenosine receptors or non-adenosine receptors, expanding its biological roles in developmental, physiological, and pathological contexts.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human brain. Anti-ADORA1 antibody was used at 1/200 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
