Background

ACVR1B (activin A receptor type 1B, also known as ALK4) is a type I transmembrane serine/threonine kinase receptor encoded by the ACVR1B gene. The human protein is approximately 505 amino acids long with a molecular weight of about 57 kDa, and the gene is located at chromosome 12q13.13. As a key type I receptor in TGF-β superfamily signal transduction, ACVR1B mainly mediates activin A signaling: after the ligand binds the type II receptor (ACVR2A/ACVR2B), ACVR1B is recruited, the type II receptor phosphorylates its GS domain and activates its kinase activity, which in turn phosphorylates SMAD2/3 to initiate the canonical signaling pathway. In tumor biology, ACVR1B is primarily regarded as a tumor suppressor gene, and its inactivating mutations are found in pancreatic cancer and ER-negative breast cancer; knockdown of ACVR1B can promote pancreatic cancer cell growth and tumorigenesis. Mechanistically, loss of ALK4 increases cell-surface stability of TGF-β receptors by altering their glycosylation, thereby paradoxically enhancing TGF-β signaling and promoting epithelial–mesenchymal transition and metastasis. In addition, ACVR1B is involved in the regulation of skeletal muscle atrophy and regeneration; its genetic variants are associated with pituitary tumors, and in systemic sclerosis it is implicated in activin/ACVR1B signaling-mediated collagen production.
