Background
ACSS3 (Acyl-CoA Synthetase Short-Chain Family Member 3) is also localized in the mitochondrial matrix and is encoded by the ACSS3 gene on chromosome 12q21.31. Functionally, ACSS3 catalyzes the synthesis of acetyl-CoA from short-chain fatty acids. In contrast to ACSS1, which prefers acetate, ACSS3's preferred substrate is propionate, with much lower affinity for acetate and butyrate. This enzyme is involved in the ketone body biosynthetic process.
ACSS3 exhibits a complex, bidirectional role in cancer. In prostate cancer (PCa), ACSS3 is downregulated due to promoter methylation, and its low expression predicts poor prognosis. Restoring ACSS3 expression reduces lipid droplet (LD) deposits by downregulating the LD coat protein PLIN3, thereby increasing ER stress, promoting apoptosis, and inhibiting the progression of castration-resistant prostate cancer (CRPC). Conversely, in glioma, non-small cell lung cancer, and ovarian cancer, ACSS3 is highly expressed and is associated with promoting tumorigenesis and malignant progression. This disparity suggests that ACSS3's function is highly tissue/tumor type-specific.
