Background
Acetyl-alpha-tubulin (Lys40) refers to a specific protein epitope formed when the lysine residue at position 40 of alpha-tubulin undergoes acetylation, and it is one of the most extensively studied post-translational modifications of microtubules. This modification is added by the acetyltransferase αTAT1 and removed by deacetylases such as HDAC6 and Sirtuin 2. In terms of localization, Lys40 is located on the inner surface of the microtubule lumen rather than on the outer surface, and this unique spatial position makes it difficult for microtubule-associated proteins or motor proteins to access it directly. Functionally, K40 acetylation does not simply "mark" stable microtubules; rather, it increases microtubule flexibility by weakening lateral interactions between protofilaments, thereby helping long-lived microtubules resist structural damage caused by repeated mechanical stress and playing a protective role against "mechanical aging." In experimental research, site-specific antibodies against this residue (such as clones RM318 or D20G3) are commonly used in Western blotting and immunocytochemistry to detect and quantify the acetylation level of alpha-tubulin K40, and they have been validated to show no cross-reactivity with the non-acetylated form of alpha-tubulin.

Immunocytochemistry
ICC analysis of HeLa cells treated with TSA(1uM,16h) (top panel) and untreated HeLa cells (below panel). Anti- Acetyl-alpha-Tubulin (Lys40) antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
Immunofluorescence
IF shows positive staining in paraffin-embedded rat testis. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
