Background

NeuroD1 (Neurogenic Differentiation 1) is a basic helix-loop-helix (bHLH) transcription factor that serves as a master regulator in the development of the nervous system and pancreas, primarily functioning to commit progenitor cells to a neuronal fate by activating downstream neurogenic genes while simultaneously suppressing glial differentiation pathways. Expressed transiently during embryogenesis in specific regions of the central and peripheral nervous systems, NeuroD1 binds to E-box DNA sequences as a heterodimer with E-proteins to drive the expression of targets essential for neuronal maturation, migration, and survival, making it critical for the formation of structures such as the hippocampus, cerebellum, and retina; beyond its developmental role, it is also pivotal in pancreatic beta-cell differentiation and has been identified as a potent reprogramming factor capable of directly converting non-neuronal cells, including astrocytes and fibroblasts, into functional neurons, thereby holding significant therapeutic potential for treating neurodegenerative diseases and repairing neural injury through endogenous regeneration or cell replacement strategies.
