Flow cytometric analysis of 50ng/ml PMA, 1μg/ml Ionomycin and 1μg/ml BFA cultured human PBMC (human peripheral blood mononuclear cell) labelling human IL-9 antibody at 1/2000 dilution (0.1 μg) / (Right Panel) compared with a Mouse IgG2b, κ Isotype control (Left Panel). Goat Anti - Mouse IgG Alexa Fluor® 647 was used as the secondary antibody. Then cells were stained with CD3 - Alexa Fluor® 488 separately. Gated on total viable cells.

Background
Interleukin-9 (IL-9) is a 14 kDa glycoprotein cytokine primarily produced by activated Th2, Th9, and innate lymphoid cells that signals through a heterodimeric receptor composed of the IL-9Rα chain and the common γ-chain, thereby activating JAK/STAT, PI3K/AKT, and MAPK pathways to exert pleiotropic effects on hematopoietic and epithelial cells, promoting mast-cell proliferation and differentiation, enhancing mucus production by goblet cells, modulating B-cell responses, and contributing to the pathogenesis of allergic asthma, atopic dermatitis, and certain hematologic malignancies, while also exerting context-dependent anti-tumor or pro-tumor activities depending on the cellular milieu.
