Flow cytometric analysis of Human CD158b expression on human peripheral blood Leukocytes. Human peripheral blood Leukocytes were stained with PE Mouse Anti-Human CD56 Antibody and either APC Mouse IgG2a, κ Isotype Control (Left panel) or SDT APC Mouse Anti-Human CD158b/j Antibody (Right panel) at 5 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD158b/j is a collective designation for the killer-cell immunoglobulin-like receptors (KIR2DL2, KIR2DL3 and KIR2DS2), 50–58 kDa type-I transmembrane glycoproteins expressed chiefly on natural killer cells and minor T-cell subsets; each molecule carries two extracellular Ig-like C2 domains that recognize distinct HLA-C alleles (HLA-Cw1/3/7 for the inhibitory forms), and while the long-tailed CD158b variants (KIR2DL2/L3) deliver inhibitory signals via dual cytoplasmic ITIM motifs to prevent target-cell lysis, the short-tailed CD158j variant (KIR2DS2) can trigger activation, thereby collectively fine-tuning innate cytotoxic responses and influencing immune escape in infection, transplantation and cancer.
