Background

Androgen receptor (AR), also known as ANDR, is a ligand-activated nuclear transcription factor whose structure comprises an N-terminal transactivation domain, a DNA-binding domain, a hinge region, and a C-terminal ligand-binding domain. As the central mediator of the androgen signaling pathway, AR undergoes dimerization upon ligand binding and translocates into the nucleus, where it regulates target gene transcription, thereby driving male sexual differentiation as well as the growth and survival of prostate cells. Clinically, AR is a key driver of the development and progression of prostate cancer, and the vast majority of prostate cancers depend on AR signaling for growth; therefore, androgen deprivation therapy and AR antagonists constitute the cornerstone of treatment. However, AR gene amplification, mutations, or splice variants (such as AR-V7) can lead to therapeutic resistance, making AR an important target in research aimed at overcoming resistance and in the development of novel drugs.

Bioactivity

The ANDR(LBD) activity was detected using HTRF binding assay. The reaction was performed by incubating the ANDR(LBD) protein, BRD4 and HLD-0915 at room temperature, then reading ratio 665/620 with BMG.
