Background

TBR2/Eomes is a T-box transcription factor that plays crucial roles in various developmental processes. It is preferentially enriched in intermediate progenitors and supports cortical neurogenesis expansion. In the developing cerebral cortex, it is specifically expressed in intermediate progenitors (IPs), which are derived from radial glia progenitors and generate glutamatergic projection neurons. TBR2/Eomes is essential for the differentiation and subtype specification of these neurons, and it regulates the transcriptome by activating or repressing hundreds of direct target genes. Additionally, TBR2/Eomes is involved in the formation and maintenance of certain retinal ganglion cells. It also plays a role in the maintenance of the trophectoderm lineage by sustaining chromatin accessibility and promoting gene regulatory networks.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human tonsil. Anti-TBR2/Eomes antibody was used at 1/1000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Negative control: IHC shows negative staining in paraffin-embedded human kidney.

Immunofluorescence

IF shows positive staining in paraffin-embedded mouse E14 embryo. Anti-TBR2/Eomes antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
