Concept
The Ubiquitin-Specific Protease (USP) family is the largest and most versatile subclass of deubiquitinases (DUBs) in eukaryotes, with approximately 60 human isoforms. USPs function as cysteine proteases that remove ubiquitin chains from target proteins, thereby modulating protein stability, localization, activity, and protein–protein interactions. All USPs contain a conserved catalytic triad (Cys-His-Asp/Asn) but are distinguished by diverse auxiliary domains—including UBA, UBL, ZnF, TRAF, and RNA-binding motifs—that dictate substrate specificity, subcellular localization, and regulatory dynamics. USPs govern nearly all cellular processes, making them central to health and disease.
