Selective Chemical Labeling of Protein Lysine Methacrylation
1. Literature Information
This work reveals a photocatalytic thiol–Michael addition strategy for highly specific chemical labeling and proteomic identification of lysine methacrylation, a newly discovered acylation modification highly similar to crotonylation.
2. Research Background
Lysine methacrylation is a structural isomer of crotonylation, widely distributed in histones and related to mitochondrial metabolism and genetic diseases. Traditional antibody tools cannot distinguish methacrylation from crotonylation due to high structural similarity, causing severe crossinterference and hindering functional study. There is an urgent need for chemical biology methods to achieve selective labeling and enrichment.
3. Research Approach
Based on thermodynamic and steric differences between methacrylamide and crotonamide, researchers designed a watersoluble probe with benzene thiol reactive group and azide handle. Under photocatalysis, thiol radicals selectively add to methacrylamide double bonds with minimal reaction to crotonamide. Labeled proteins are biotinylated via click chemistry, enriched by streptavidin beads and identified by highresolution mass spectrometry.
4. Research Outcomes
· The probe achieves over 85% selectivity for methacrylation even under high crotonylation background.
· This workflow enables systematic identification of novel methacrylation sites on histones and nonhistone proteins.
· Multiple new substrates involved in chromatin regulation, transcription and cell cycle are discovered, expanding the methacrylation regulatory network.
· The method overcomes limitations of antibodybased enrichment and enables unbiased proteomewide profiling.
