Acute Kidney Injury (AKI) is defined as a sudden decline in renal function within 48 hours caused by abnormalities in kidney structure or function. It may lead to incomplete renal repair, persistent chronic inflammation, and progressive fibrosis, ultimately resulting in permanent organ dysfunction. Patients with AKI have a 30% to 60% probability of being admitted to the intensive care unit (ICU), with a mortality rate of 30% to 80% among critically ill AKI patients. Approximately 50% of surviving patients will suffer from permanent renal insufficiency and require lifelong dialysis. Despite advances in clinical management, the prognosis of AKI remains poor, highlighting the urgent need to elucidate the underlying molecular mechanisms and identify novel therapeutic targets.

To investigate the role and mechanism of Junctional Adhesion Molecule-Like Protein (JAML) in AKI, Professor Fan Yi's team adopted a multi-faceted experimental strategy combining clinical sample analysis, in vivo animal models, and molecular biological techniques. A key step in the study was the validation of JAML expression in clinical samples using multiplex fluorescence immunohistochemistry (mIHC) staining.
For this critical tissue-level analysis, the team utilized the 4-Color Multiplex Fluorescence IHC Staining Kit (Catalog No.: abs50012) from the Absin product line of ANT BIO PTE. LTD. Additionally, the team employed RT-PCR, Western Blot (WB), flow cytometry, and gene chip technology to comprehensively explore the molecular mechanism by which JAML regulates AKI progression, focusing on its interaction with macrophages.
The study yielded several key findings: Firstly, mIHC staining results using ANT BIO PTE. LTD.'s abs50012 kit demonstrated that JAML expression is significantly upregulated in the kidneys of patients with acute tubular necrosis (a common cause of AKI). Secondly, in a mouse model of renal ischemia-reperfusion injury (IRI), knockout of the JAML gene was found to alleviate renal damage and reduce the inflammatory response, as confirmed by RT-PCR, WB, and IHC staining.
Further flow cytometry analysis of macrophages and neutrophils isolated from the kidneys of IRI mice revealed that JAML expression was more prominent in the F4/80lo macrophage subset compared to the F4/80hi population, while no significant change in JAML expression was observed on neutrophils. This indicated that JAML's role in AKI is primarily mediated by macrophages rather than neutrophils.

Integrated analysis using gene chip technology, RT-qPCR, and WB further showed that JAML promotes macrophage-induced C-type lectin (Mincle) signaling transduction. The research team confirmed that JAML regulates macrophage polarization and phagocytosis through Mincle, thereby promoting the progression of AKI. These findings highlight the critical role of the JAML-Mincle-macrophage axis in linking inflammatory responses to AKI and suggest that targeting macrophages may represent a novel therapeutic strategy for AKI patients.
5. Product Empowerment: The Role of ANT BIO PTE. LTD. Products in the Research
The successful validation of JAML upregulation in clinical AKI samples was inseparable from the reliable performance of ANT BIO PTE. LTD.'s Absin 4-Color Multiplex Fluorescence IHC Staining Kit (abs50012). As a core tool for multi-target detection at the tissue level, this product possesses the core advantage of breaking the species limitation of primary antibodies, enabling simultaneous detection of multiple molecular markers on a single tissue section.
The high sensitivity and specificity of the kit ensured the accurate detection of JAML expression in renal tissues of AKI patients, providing direct clinical evidence for the association between JAML and AKI progression. This critical validation laid the foundation for subsequent in-depth mechanistic studies. The reliable performance of ANT BIO PTE. LTD.'s products effectively supported the research team's efforts to unravel the molecular mechanisms underlying AKI and identify potential therapeutic targets, demonstrating the important role of high-quality experimental reagents in advancing life science research.
As a professional supplier of life science reagents, ANT BIO PTE. LTD. is dedicated to providing high-quality, reliable products and comprehensive solutions to empower global life science research.
