In Vivo Immune Checkpoint Antibodies: Separating Pathway Effects from Fc-Mediated Effects

Checkpoint blockade works, which is precisely why in vivo studies of it are so easy to misread. A tumour shrinks after treatment. The question is whether the shrinkage came from blocking PD-1, from Fc-mediated killing of the cells that carry PD-1, or from an immune reaction to the injected protein. Standard reagents cannot separate those explanations. Reagents engineered to remove the Fc contribution can. This article explains the design logic and what to verify before dosing animals.

Two Arms of Checkpoint Control

T cell activation requires a recognised antigen and a second signal. Checkpoints govern that second signal in both directions. Co-stimulatory molecules such as CD28, ICOS and 4-1BB, also called CD137, amplify activation, proliferation, differentiation and survival. They act as an accelerator.

Co-inhibitory molecules such as CTLA-4, also called CD152, PD-1, also called CD279, and LAG-3 transmit the opposite instruction. They are upregulated after activation and limit its duration and intensity. This balance prevents autoimmune damage and excessive inflammation. It also creates the opportunity that tumours exploit.

How Tumours Take Control of the Brake

Tumour cells and their neighbours overexpress ligands for the inhibitory arm. PD-L1 is the best known, and CD80 or CD86 serve CTLA-4. Myeloid-derived suppressor cells and tumour-associated macrophages contribute to the same ligand pool.

When tumour-specific T cells arrive, their inhibitory receptors meet an excess of ligand. The result is functional exhaustion: proliferation stalls, cytokine output falls, and apoptosis may follow. The tumour has not defeated the T cell directly. It has persuaded the T cell to stand down. Blocking the interaction reverses that instruction.

Why CTLA-4 and PD-1 Act in Different Places

The two approved classes are not interchangeable, and their anatomy explains why. Anti-CTLA-4 antibodies act mainly during the priming phase in lymph nodes. They block CTLA-4 binding to B7 molecules and weaken early restraint on T cell activation. More effector cells are generated and released.

Anti-PD-1 and anti-PD-L1 antibodies act mainly during the effector phase inside the tumour microenvironment. They interrupt PD-1 engagement with PD-L1 or PD-L2 and reverse the exhausted state locally. One expands the repertoire; the other restores function in situ. Combination logic follows directly from that division of labour.

Newer targets including LAG-3, TIGIT, TIM-3 and VISTA are in clinical development. The aim is to overcome resistance to existing agents and to widen the responding population.

The Fc Question That Decides Interpretation

An IgG1 antibody can recruit effector functions. Fc receptors on natural killer cells and macrophages trigger antibody-dependent cellular cytotoxicity, and complement triggers complement-dependent cytotoxicity. In a therapeutic setting those effects may help. In a mechanism study they contaminate the result.

A D265A substitution in the Fc region removes Fc receptor binding and eliminates both activities. What remains is pure blockade of the PD-1 pathway. If a phenotype disappears when the Fc-silent version is used, the phenotype was effector-mediated. If it persists, the pathway itself is responsible. That comparison is the whole point of the design.

Proving Bioactivity Before Dosing

Binding affinity is necessary but not sufficient. Two functional assays carry most of the weight. A cell-based reporter assay confirms that the antibody interrupts PD-1 signalling in a measurable, dose-dependent way. A primary T cell activation assay confirms the same effect in a more physiological setting, where PD-L1 or PD-L2 is supplied by a presenting cell.

Both assays should be run alongside an isotype control that matches the host species and the Fc format. Without that control, a modest increase in T cell activity cannot be attributed to blockade rather than to general perturbation of the culture.

Purity and Endotoxin in Repeated Dosing Studies

An in vivo study usually involves repeated administration over weeks. Every impurity is therefore administered repeatedly as well. Endotoxin is the most consequential. Levels below 1.0 EU per milligram are the working expectation for reagents intended for repeated mouse dosing.

Three further specifications deserve attention. Host protein residues indicate how clean the expression and purification process is. Nucleic acid residues carry a similar meaning. Sterility protects against confounding infection in immunocompromised models. A supplier that reports these values is describing a process, not just a product.

Matching the Model to the Question

The model choice constrains what the antibody can show. Syngeneic tumour implants in immunocompetent mice allow a complete immune response and are the standard for efficacy work. Humanized PDX models allow human tumour tissue but require a compatible host immune system. Genetically engineered models permit defined lesions and temporal control.

Disease context changes the readout further. Experimental autoimmune encephalomyelitis and collagen-induced arthritis test the PD-1 axis in autoimmunity, where blockade may worsen disease. Chronic lymphocytic choriomeningitis virus infection tests whether blockade reverses exhaustion and restores antiviral function. Each setting uses the same reagent to answer a different question.

The set supports ligand-side and receptor-side blockade in one model. That pairing is useful when a result must be attributed to the pathway rather than to a single molecule. The CD4 reagent serves as a depletion control, a role that is often missing from published combination studies.

Selection Notes

Four checks separate a usable in vivo reagent from a costly variable. Confirm the Fc format and whether effector function is intact. Confirm endotoxin values per milligram, not per vial. Confirm that potency was measured functionally, not only by binding. Confirm that an isotype control from the same production platform is available.

A study that controls all four can attribute an observed phenotype to the pathway. A study that controls none may still produce a publishable result, but the mechanism claim will remain open to challenge. All products described here are supplied for research use only.
