Background
Stat4 (signal transducer and activator of transcription 4) is an important member of the STAT protein family, encoded by the STAT4 gene located on human chromosome 2, and plays a central regulatory role in the immune system. This protein possesses a typical STAT family structure, containing key functional domains including a DNA-binding domain and an SH2 domain. The primary function of STAT4 is to mediate interleukin-12 (IL-12) signaling; upon IL-12 stimulation, it is phosphorylated and activated by JAK kinases, subsequently forming homodimers and translocating into the nucleus to initiate the transcription of target genes such as interferon-gamma (IFN-γ), thereby driving the differentiation of naive CD4+ T cells into Th1 cells, which is critical for defense against intracellular pathogen infections. In addition, STAT4 also plays an indispensable role in the functional regulation of various immune cells including NK cells and dendritic cells. Its functional abnormalities are closely associated with multiple autoimmune diseases, and STAT4 gene polymorphisms (such as the rs7574865 locus) have been confirmed to significantly increase susceptibility to rheumatoid arthritis and systemic lupus erythematosus; meanwhile, its role in tumor development is also receiving increasing attention, with its high expression in colorectal cancer being positively correlated with tumor invasion depth.
