Background
RBPMS (RNA-binding protein with multiple splicing) is a vertebrate-conserved RNA-binding protein that harbors a single, ~80-aa RNA-recognition motif (RRM) flanked by N- and C-terminal extensions rich in prolines, is expressed as numerous mRNA and protein isoforms from a >230-kb locus, and functions as a master post-transcriptional regulator by directing alternative splicing, stability and translation of target mRNAs critical for cardiac, smooth-muscle, retinal ganglion cell and oocyte development, as well as for DNA-damage responses through interactions with RIF1 and modulation of TGF-β/BMP signaling pathways.

Immunocytochemistry
ICC shows positive staining in A549 cells. Anti- RBPMS antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
Immunofluorescence
IF shows positive staining in paraffin-embedded mouse retina. Anti- RBPMS antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
