Description

Testing Principle:
This kit uses double antibody sandwich ELISA.Specific anti-rat IFN-γ Capture Antibody is precoated on a high affinity conjugate plate. Standard and test samples were added successively into the well of the plate, and the samples were fully shaken and mixed and placed at room temperature for 2-hour incubation. The IFN-γ in the samples bound to the solid phase antibody. Wash thoroughly to remove free and unbound components, then add a biotinylated detection antibody and incubate. After washing again, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is added. After washing, signal enhancer was added for incubation. After washing and removing unbound substances, SA-HRP was added again. After washing again, TMB color substrate was added and incubated in dark at room temperature for color development. The depth of color reaction was positively correlated with the concentration of IFN-γ in the sample. The termination solution was added to terminate the reaction, and the absorbance value was measured at 450 nm detection wavelength (correction wavelength 570-630 nm) using a microplate analyzer.

Interferon γ (IFN-γ), a soluble dimer cytokine, is the only member of type II interferon.It is secreted by natural killer cells (NK) and natural killer T cells (NKT) and plays a role in innate immunity.It is secreted by CD4Th1 and CD8 cytotoxic T cells during antigen-specific immunity.IFN-γ or type II interferon plays an important role in innate A immunity and adaptive immunity against viral and certain bacterial and protozoan infections.IFN-γ is an important activator of macrophages and an inducer of type II major histocompatibility complex (MHCII) expression.Abnormal expression of IFN-γ is associated with many autoinflammatory and autoimmune diseases.In addition to direct inhibition of viral replication, IFN-γ is important to the immune system in terms of its immune-stimulating and immune-regulatory functions.
IFN-γ is a marker of disease in many different pathological conditions, including infections, autoimmune diseases, transplant rejection, allergic reactions, and diabetes mellitus.
