Background
Podocalyxin-like protein 1 (PODXL) is a type I transmembrane protein belonging to the CD34 family of sialomucins, typically expressed in kidney podocytes, vascular endothelial cells, hematopoietic progenitors, mesothelium, and certain neurons. It has a highly glycosylated extracellular domain, a single transmembrane domain, and a short cytoplasmic tail. PODXL functions primarily as an anti-adhesive molecule, regulating cell morphology and adhesion, and is crucial for kidney development and function. In cancer, PODXL is often overexpressed and associated with aggressive tumor phenotypes, poor prognosis, and potentially chemoresistance.
