Background

Phospho-Src Family (Tyr416) refers to the Src family of protein tyrosine kinases when phosphorylated at the tyrosine residue 416. This post-translational modification is crucial for the activation of Src kinases, which include Src, Lyn, Fyn, Yes, Lck, Blk, and Hck. Phosphorylation at Tyr416, located within the activation loop of the kinase domain, upregulates enzyme activity, while phosphorylation at another regulatory site, Tyr527, downregulates it. In the context of cancer, Src activity is often dysregulated, contributing to tumorigenesis and cancer progression. Understanding the mechanisms of Src activation, such as phosphorylation at Tyr416, is important for developing targeted therapies against cancer.

Immunocytochemistry

ICC analysis of A431 cells treated with pervanadate (50mM, 5min) (top panel) and A431 cells untreated with pervanadate (50mM, 5min) (below panel). Anti- Phospho-Src Family (Tyr416) antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
