Background

NTSR1 (neurotensin receptor type 1) is a high-affinity G protein-coupled receptor (GPCR) encoded by the NTSR1 gene, belonging to the seven-transmembrane protein family, with its gene localized to the q13.33 region of human chromosome 20. This protein is the primary effector receptor for the neuropeptide neurotensin (NTS), mediating various physiological functions through NTS binding, including blood pressure regulation, thermoregulation, antinociception (analgesia), feeding behavior, as well as intestinal motility and secretion. At the level of signal transduction, upon binding with NTS, NTSR1 activates multiple downstream signaling pathways, including PLC-mediated intracellular calcium release, the PKC/RAF-1/MAPK-ERK1/2 cascade, and the PI3K/AKT pathway, and the activation of these signaling networks in turn regulates cell proliferation, survival, migration, and cytoskeletal dynamics. Precisely because of its strong pro-proliferative and pro-migratory capabilities, NTSR1 plays a tumor-promoting role in various malignancies, with its expression being upregulated in multiple tumors including glioma, pancreatic ductal adenocarcinoma, gastric cancer, and prostate cancer, and is closely associated with poor patient prognosis and enhanced tumor aggressiveness. In addition, NTSR1 is also genetically linked to central nervous system disorders such as schizophrenia, drug addiction, and Parkinson's disease, and its functional abnormalities also involve susceptibility to alcohol and opioid dependence. In recent years, three-dimensional structures of NTSR1 in complex with G proteins have been resolved using techniques such as cryo-electron microscopy, providing an important foundation for in-depth understanding of its allosteric regulatory mechanisms and biased signal transduction (such as biasing toward the β-arrestin pathway to attenuate psychostimulant effects), making it a hotspot target in drug development and tumor-targeted therapy.
