Flow cytometric analysis of human mDCs (mature human monocyte‑derived dendritic cells) fixed with 4% paraformaldehyde and permeabilized with 0.1% Tween. Cells were labeled with Mouse Anti-Human CD208 antibody at 1/200 dilution (1 μg) / (right panel) compared with a Mouse IgG1, κ Isotype Control / (left panel). Goat Anti-Mouse IgG Alexa Fluor® 647 was used as the secondary antibody. Then cells were stained with CD11c - Brilliant Violet 711™ antibody separately. Total viable cells, as determined by Fixable Viability Dye 452 (S0B88801), were used for analysis. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.

Background
CD208, also known as DC-LAMP or LAMP-3, is a member of the lysosome-associated membrane protein (LAMP) family. It was first identified as a specific marker for mature dendritic cells (DCs), with its expression significantly upregulated during DC differentiation and maturation. As a lysosomal membrane glycoprotein, its structure contains a conserved luminal domain with a novel β-prism fold, a feature that allows it to play an important role in the architecture of the lysosomal glycocalyx. Its functions extend far beyond serving as a mere marker: CD208 is involved in the unfolded protein response (UPR) and autophagy, and participates in the fusion of lysosomes with autophagosomes, thereby influencing cellular protein degradation and survival. Furthermore, CD208 expression is not restricted to activated dendritic cells; it is also present in normal alveolar type II epithelial cells (pneumocytes) and colocalizes with intracellular MHC class II molecules, suggesting a potential role in antigen presentation. Therefore, CD208 holds significant importance in immune responses, regulation of lysosomal function, and the physiological and pathological states of specific tissues, such as the lungs.
