Background
IL-27 p28 (also known as IL-27A) is a unique subunit of the heterodimeric cytokine IL-27, which covalently links with another subunit, EBI3 (Epstein-Barr virus-induced gene 3), via a disulfide bond to form the biologically active IL-27 complex. Structurally, p28 belongs to the IL-6/IL-12 family of α-helical four-helix bundle cytokines and is primarily produced by activated antigen-presenting cells (such as dendritic cells and macrophages) following stimulation with Toll-like receptor agonists or interferons. The function of p28 exhibits significant cell type-dependent effects: in naive T cells, IL-27 composed of p28 primarily activates STAT1 and STAT3 signaling pathways through the gp130 and WSX-1 receptor complex, promoting Th1-type immune responses and enhancing the cytotoxic function of CD8⁺ T cells, thereby playing a critical role in anti-infective immunity. However, in regulatory T cells (Tregs), p28 can signal in a non-canonical manner independently of EBI3, and IL-27 itself also exhibits immunosuppressive properties, capable of limiting excessive inflammation and inducing IL-10 production. Consequently, it plays a dual pro-inflammatory and anti-inflammatory role in autoimmune diseases and antitumor immunity, positioning it as a double-edged sword target of great interest in the field of immunotherapy.
