Background

Proprotein convertase subtilisin/kexin type 9 (PCSK9) is a key regulator of cholesterol metabolism, primarily known for its role in degrading low-density lipoprotein receptors (LDLR) in the liver. By binding to LDLR and promoting its lysosomal degradation, PCSK9 reduces the liver's ability to clear circulating LDL cholesterol (LDL-C), leading to elevated plasma LDL-C levels. Gain-of-function mutations in the PCSK9 gene are associated with autosomal dominant hypercholesterolemia, while loss-of-function variants correlate with reduced LDL-C and cardiovascular risk.

Linearity
The concentrations of Human PCSK9 were measured and interpolated from the target standard curves and corrected for sample dilution.
Sample is undiluted samples are as follows: HepG2 human hepatocellular carcinoma cells culture supernatant (5%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 25.61 ng/mL in HepG2 cells culture supernatant.
