Background
Interleukin-18 (IL-18), a proinflammatory cytokine primarily produced by macrophages, plays a complex role in immune regulation by potently inducing IFN-γ production and, in synergy with IL-12, shifting B-cell responses away from IgE. Its activity is activated by caspase-1 and inhibited by IL-18 binding protein. Paradoxically, IL-18 can also promote IgE production in certain contexts, highlighting its dual function in allergic diseases, while in rheumatoid arthritis, it synergizes with IL-12 and IL-15 to sustain pathogenic Th1 responses and monokine production, underscoring its significance as a therapeutic target.

Linearity
The concentrations of Human IL-18 were measured and interpolated from the target standard curves and corrected for sample dilution.
The sample is undiluted samples are as follows: human serum (50%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 1916.26 pg/mL in human serum.

To assess the linearity of the assay, two samples were spiked with high concentrations of Human IL-18 in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
