Background
Human Growth Differentiation Factor 15 (GDF-15), also known as MIC-1 or PDF, is a stress-responsive cytokine belonging to the TGF-β superfamily. Primarily secreted by macrophages, epithelial cells, and placental tissues under pathological conditions, this 25-kDa dimeric protein signals through the glial cell-derived neurotrophic factor (GDNF) family receptor α-like (GFRAL)-RET complex in the brainstem. It regulates systemic energy homeostasis by suppressing appetite and modulating insulin sensitivity, while also functioning as a biomarker for cellular stress, cardiovascular disease, and cancer progression. Elevated GDF-15 levels correlate with cachexia, heart failure, and tumor metastasis, making it a therapeutic target for metabolic disorders and a diagnostic indicator for tissue damage. Its expression increases >100-fold during starvation, inflammation, and oxidative stress.

The concentrations of Human GDF-15 were measured and interpolated from the target standard curves and corrected for sample dilution.
#1 sample is undiluted samples are as follows: HepG2 Cells culture was 3×10^2fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 87.32 ng/mL in HepG2 Cells culture
To assess the linearity of the assay, The samples was spiked with high concentrations of Human GDF-15 in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
