Background
HECTD4 (HECT domain E3 ubiquitin protein ligase 4) is a protein encoded by the HECTD4 gene, which functions primarily as an E3 ubiquitin ligase that marks specific target proteins for degradation or functional regulation by attaching ubiquitin molecules to them. In both physiological and pathological processes, HECTD4 plays significant roles: on one hand, biallelic mutations in this gene have been identified as the cause of an autosomal recessive neurodevelopmental disorder characterized by epilepsy, spasticity, and agenesis or hypoplasia of the corpus callosum; on the other hand, studies have revealed its complex involvement in cancer, where it can exert tumor-suppressive effects by promoting the degradation of proteins such as COX-2, yet it may also be suppressed by certain tumor-associated non-coding RNAs (e.g., circNEIL3 and HEIH), thereby indirectly promoting tumor progression. Furthermore, single nucleotide polymorphisms in this gene have been associated with the risk of colorectal cancer.
