Background

GABRA6, also known as Gamma-aminobutyric acid type A receptor subunit alpha-6, is an important subunit of the GABA_A receptor complex, which is the ionotropic receptor for the inhibitory neurotransmitter GABA. It is encoded by the GABRA6 gene and is primarily and specifically expressed in the granule cells of the adult mammalian cerebellum, with its expression rapidly upregulated during a critical period of postnatal development, under the precise regulation of various transcriptional and epigenetic mechanisms. Functionally, the GABRA6 subunit is a core component of extrasynaptic receptors that are highly sensitive to GABA (such as α6β2/3γ2 or α6β2/3δ). These receptors exhibit slow-desensitizing current kinetics and mediate tonic inhibitory currents, thereby continuously controlling the excitability of granule cells and finely regulating cerebellar input/output functions. Furthermore, GABRA6 plays critical roles in alcohol sensitivity, benzodiazepine binding and its mediated ataxia, and cerebellar motor disorders, and its gene polymorphisms are also associated with genetic susceptibility to neuropsychiatric disorders such as Alzheimer's disease, autism, and epilepsy.

Immunocytochemistry

ICC shows positive staining in SH-SY5Y cells. Anti-GABRA6 antibody was used at 1/400 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
