Background

FRS2-α (fibroblast growth factor receptor substrate 2-α, also known as SNT1) is a critical lipid-anchored adapter protein in the FRS2 family that serves as a core mediator in downstream signal transduction of fibroblast growth factor receptors (FGFRs) and neurotrophic factor receptors (such as TRK). Its molecular structure includes a myristoylation sequence responsible for anchoring to the cell membrane, a phosphotyrosine-binding (PTB) domain that binds the juxtamembrane region of receptors, and a tyrosine-rich C-terminal region. Upon receptor activation, FRS2-α undergoes tyrosine phosphorylation, and specific tyrosine sites in its C-terminus (such as Tyr196 and Tyr436) subsequently serve as docking platforms for recruiting SH2 domain-containing signaling proteins (such as the Grb2-Sos complex and the protein tyrosine phosphatase SHP-2), thereby activating key signaling pathways such as Ras-MAPK to transduce extracellular signals to the nucleus for regulating cell growth, proliferation, and differentiation. In addition, its function is subject to negative feedback regulation mediated by MAPK, and it plays important roles in developmental biology (such as ocular development) and RET oncogene signal transduction, with broad tissue distribution but higher expression levels in organs such as the brain and kidney.
