Background

FGF4 is a secreted signaling molecule belonging to the fibroblast growth factor (FGF) family. During embryonic development, FGF4 is a key factor in maintaining the self-renewal of pluripotent stem cells and regulating the differentiation of early germ layers. It activates downstream signaling pathways such as MAPK/ERK, thereby playing decisive roles in embryo implantation, gastrulation, and limb bud development. In addition, FGF4 is also involved in angiogenesis, tissue repair, and nervous system development in adult tissues. However, aberrant expression of FGF4 is closely associated with various cancers; its gene amplification or overexpression can drive tumor cell proliferation, survival, and invasion, and is particularly regarded as an important factor promoting malignant progression and poor prognosis in breast cancer, prostate cancer, and esophageal cancer.
