Background
BMP4 (Bone Morphogenetic Protein 4) is a key member of the BMP subfamily within the transforming growth factor-β (TGF-β) superfamily. It is a highly evolutionarily conserved secreted signaling molecule that exerts pleiotropic regulatory functions in embryonic development, organogenesis, and tissue homeostasis maintenance. Its precursor protein is proteolytically cleaved to generate the mature active dimer, which exerts its effects by binding to and activating a heterotetrameric receptor complex composed of BMPR-IA, BMPR-IB, and BMPR-II, subsequently phosphorylating the downstream transcription factors Smad1/5/8. These then form complexes with Smad4 and translocate to the nucleus to regulate target gene expression, while signaling can also be transduced through non-Smad pathways (such as MAPK and PI3K pathways). During embryonic development, BMP4 is one of the earliest critical morphogens involved in establishing the body axis and determining dorsoventral polarity. In the early embryos of vertebrates, the concentration gradient of BMP4 dictates the boundary between neuroectoderm and epidermal ectoderm fates, with its activity finely regulated by antagonists such as Noggin, Chordin, and Follistatin to generate precisely patterned signals. Furthermore, BMP4 promotes osteogenic differentiation and chondrogenesis in skeletal development, and also plays irreplaceable roles in the morphogenesis of organs such as teeth, heart, kidneys, and limbs. In adult tissues, BMP4 participates in bone remodeling, adipocyte differentiation, and iron metabolism regulation. Its dysregulated expression is associated with various diseases, including fibrodysplasia ossificans progressiva (FOP), pulmonary arterial hypertension, tumorigenesis (such as colorectal cancer and prostate cancer), and congenital heart defects. Thus, BMP4 is not only a core research molecule in developmental biology but also an important therapeutic target in the fields of regenerative medicine and disease treatment.

IHC shows positive staining in paraffin-embedded human cardiac muscle. Anti-BMP4 antibody was used at 1/200 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
