Background

TPBG/5T4 (trophoblast glycoprotein/5T4 oncofetal antigen) is an approximately 72 kDa type I transmembrane glycoprotein whose extracellular domain forms a horseshoe-shaped rigid core composed of eight leucine-rich repeats (LRRs), with an intracellular domain containing a PDZ-interaction motif; functionally, it drives cell migration and tumor metastasis by inhibiting canonical Wnt/β-catenin signaling while simultaneously activating noncanonical Wnt pathways, regulating the CXCL12/CXCR4 chemotactic axis, and promoting epithelial-mesenchymal transition (EMT). Clinically, 5T4 is highly expressed in approximately 80% of solid tumors while being restricted in normal adult tissues, making it an ideal target for tumor immunotherapy that has been exploited in the development of vaccines (Trovax), antibody-drug conjugates (ADCs), CAR-T/TCR-T cell therapies, and superantigen fusion proteins (Naptumomab), and is closely associated with cancer stem cell properties and poor prognosis in multiple malignancies.
