Format

(1)Nanomagnetic beads: Dextran-coated iron oxide nanomagnetic beads coupled with anti-mouse Ly6G monoclonal antibody, stored in a PBS buffer containing BSA, Poloxamer 188, and NaN3in PBS buffer;

(2)Mouse Fc receptor blocker: Recombinant antibody, stored in a PBS buffer containing Proclin 300.

2 mL magnetic beads: suitable for 2 × 10⁹Total cell count: up to 200 sorting runs can be performed.

Principle of Separation

First, add a mouse Fc receptor blocker to the sample, then use Ly6G Nanobeads, mouse to magnetically label Ly6G⁺ cells. After labeling is complete, load the cell suspension into a separation column placed in the sorter's magnetic field. The labeled Ly6G⁺ cells are retained within the column, while the unlabeled cells flow out. At this point, the cells that have flowed out of the column constitute the negative fraction after removal of Ly6G⁺ cells. After removing the column from the magnetic field, the Ly6G⁺ cells captured by the magnet can be eluted to obtain the positive fraction.

If it is necessary to increase the purity of Ly6G+cells, the positive fraction containing Ly6G+cells can be re-applied to a second separation column for further sorting.

Background

Mouse Ly6G is a glycosylated, phosphatidylinositol-anchored cell surface protein that is typically expressed transiently during monocyte development and continuously in mature granulocytes and peripheral blood neutrophils. Ly6G nanomagnetic beads have been developed for the direct enrichment or depletion of neutrophils from mouse lymphoid tissues.

Validation Data

Using the ly6G Nanobeads kit, mouse or S/L separation columns, and a magnetic separator, Ly6G+ cells were enriched from or depleted of C57 mouse bone marrow cells. Subsequently, cells were stained with FITC Rat Anti-Mouse Ly6G Antibody (1A8) (SDT Cat: S0B1680) and APC anti-mouse/human CD11b Antibody, and live-cell populations were gated for analysis.
