Background

TCF-4 (Transcription Factor 4), a 667-amino-acid basic helix–loop–helix protein encoded by the TCF4 gene on chromosome 18q21.2, is a master regulator of neurodevelopment, immune tolerance and epithelial homeostasis that binds the canonical E-box motif 5′-CACCTG-3′ to activate or repress transcription in partnership with β-catenin in the Wnt pathway, interacts with HDACs and CBP/p300 to modulate chromatin state, and whose dosage-sensitive expression is critically linked to Pitt–Hopkins syndrome and schizophrenia while its somatic amplifications and activating mutations drive colorectal, pancreatic and breast tumorigenesis by enforcing a stem-like transcriptional program.

IHC shows positive staining in paraffin-embedded human colon cancer. Anti- TCF-4 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
