Background
LICAM (L1 cell adhesion molecule) is a 200–220 kDa transmembrane glycoprotein of the immunoglobulin superfamily that functions as a pivotal regulator of neural development—guiding neuronal migration, axon outgrowth and fasciculation via homophilic and heterophilic interactions with integrins and other adhesion receptors—and, when re-expressed in cancers such as glioblastoma, lung, endometrial and renal carcinomas, is proteolytically cleaved into soluble ectodomain and cytoplasmic fragments that activate integrin–FAK–GRB2–SOS–RAS signaling, thereby enhancing tumor-cell motility, invasion, trans-endothelial migration and metastasis and serving as an independent marker of poor prognosis and a potential therapeutic target.

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-LICAM antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
