Background

ERCC1 (Excision Repair Cross-Complementation Group 1) is a 297-amino-acid protein that, together with its obligate partner XPF (ERCC4), forms a structure-specific heterodimeric endonuclease essential for nucleotide excision repair (NER) of helix-distorting DNA lesions caused by UV light or bulky adducts and for interstrand crosslink repair; the ERCC1–XPF complex incises the damaged DNA strand on the 5′ side of the lesion, enabling excision of a ~24–32-nt oligonucleotide, and its expression level or polymorphisms (e.g., C118T, C8092A) are extensively studied as predictive biomarkers for resistance to platinum-based chemotherapy in lung, ovarian, and other solid tumors because reduced ERCC1-mediated DNA repair enhances platinum-induced cytotoxicity.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human testis. Anti-ERCC1 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
