Background
IL-12 is a key immunoregulatory cytokine produced primarily by immune cells like macrophages and dendritic cells. Its fundamental role is to bridge innate and adaptive immunity. It potently stimulates the activation of natural killer (NK) cells and T cells. A major function is driving the differentiation of naive T cells into the T-helper 1 (Th1) type. This action prompts T and NK cells to produce interferon-gamma (IFN-γ), a critical cytokine for cell-mediated immunity. Consequently, IL-12 is essential for defending against intracellular pathogens and tumors. It also enhances the cytotoxic activity of immune cells, enabling them to destroy infected or malignant targets. Due to its potent effects, its activity is tightly regulated within the body. Dysregulation of IL-12 is linked to autoimmune diseases, highlighting its powerful inflammatory potential. In summary, IL-12 is a master regulator of pro-inflammatory immune responses.

Within the detection range, the human IL-12p70 protein was added to different matrices at high, medium, low concentrations.
The matrices used in these experiments were diluted 2-fold with the assay buffer before the addition of the cytokine proteins.
The results were compared with those of samples that had the same concentration of cytokines added in the assay buffer.
Quantifiction of human PBMC
Stimulated sample: Human peripheral blood mononuclear cells (PBMCs) were stimulated with 5 μg/mL PHA for 24h, and the concentration of IL-12p70 in the supernatant was measured to be 29.68 pg/mL.
Unstimulated sample: Human PBMCs were cultured for 24h, and the concentration of IL-12p70 in the supernatant was measured to be 0.25 pg/mL.
Linearity
The high-concentration human IL-12p70 protein was serially diluted using plasma, serum and cell culture supernatant matrices respectively.
The matrices used in these experiments were diluted 2-fold with the assay buffer before the addition of the cytokine proteins.
Then, the concentration of human IL-12p70 was determined and interpolated based on the target standard curve, and the sample dilution was corrected.
