Flow cytometric analysis of Mouse CD14 expression on RAW 264.7 Cells from the RAW 264.7 (Mouse Abelson murine leukemia virus-induced tumor macrophage, Right) or Neuro-2a (Mouse neuroblastoma neuroblast, Left) was stained with Pacific Blue Rat IgG2a, κ Isotype Control (Black line histogram) and SDT Pacific Blue Rat Anti-Mouse CD14 Antibody (Red line histogram) at 5 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD14 is a 375-amino-acon, pattern-recognition glycoprotein that circulates in plasma as a soluble 55 kDa fragment (sCD14) shed by GPI-anchor cleavage or exists as a 55 kDa, glycosylphosphatidylinositol-anchored membrane receptor (mCD14) on myeloid cells; in both forms it acts as the critical co-receptor for Toll-like receptor 4/MD-2, enabling innate immune detection of lipopolysaccharide and other bacterial lipoglycans, thereby triggering NF-κB–driven cytokine storms, and its serum concentration rises acutely in sepsis, making it both a biomarker and a therapeutic target for endotoxin-driven inflammation.
