Flow cytometric analysis of IL-9 expression in human PBMC polarized with Th2 Polarization Kit, Human (UA090018). Th2 polarized human PBMC were harvested and restimulated for 5 hr with PMA and Ionomycin in the presence of Brefeldin A (Right Panel) or unstimulated (Left Panel).
The cells were harvested and fixed with 4% PFA and permeabilized with Intracellular Fixation & Permeabilization Buffer Set. The cells were then stained with SDT Pacific Blue Mouse Anti-Human IL-9 Antibody at 5μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software."

Background
Interleukin-9 (IL-9) is a 14 kDa glycoprotein cytokine primarily produced by activated Th2, Th9, and innate lymphoid cells that signals through a heterodimeric receptor composed of the IL-9Rα chain and the common γ-chain, thereby activating JAK/STAT, PI3K/AKT, and MAPK pathways to exert pleiotropic effects on hematopoietic and epithelial cells, promoting mast-cell proliferation and differentiation, enhancing mucus production by goblet cells, modulating B-cell responses, and contributing to the pathogenesis of allergic asthma, atopic dermatitis, and certain hematologic malignancies, while also exerting context-dependent anti-tumor or pro-tumor activities depending on the cellular milieu.
