Background
CD337, also known as NKp30 or NCR3, is a 30 kDa type-I transmembrane glycoprotein expressed almost exclusively by resting and activated natural killer (NK) cells; it contains a single V-type immunoglobulin-like extracellular domain, a positively charged transmembrane arginine that couples to the ITAM-bearing adaptors CD3ζ and FcεRγ, and exists as six alternatively spliced isoforms that differ in signalling potency. Acting as a major activating receptor within the natural cytotoxicity receptor (NCR) family, NKp30 triggers NK-cell cytotoxicity and cytokine release against tumour, virus-infected or immature dendritic cells that have down-regulated MHC-I, by recognising ligands such as the tumour-derived nuclear chaperone B7-H6 and BAT3; however, soluble forms of these ligands can down-regulate NKp30 and foster immune evasion.
