Background

CD126, also known as the interleukin-6 receptor alpha chain (IL-6Rα), is a type I cytokine receptor that plays a crucial role in regulating cell growth and differentiation in various tissues. It is a subunit of the IL-6 receptor complex, which also includes the signal-transducing subunit glycoprotein 130 (gp130 or CD130). CD126 is primarily expressed on B cells, epithelial cells, and hepatocytes. The binding of IL-6 to CD126 forms a complex that, in association with CD130, initiates signaling pathways involved in immune responses and acute phase reactions. Dysregulated production of IL-6 and CD126 is implicated in the pathogenesis of several diseases, including multiple myeloma, autoimmune diseases, and certain cancers. Additionally, CD126 is being explored as a potential therapeutic target for CAR-T cell therapy due to its expression in various tumors.
